Melanotan I is studied primarily in melanocortin-1 receptor signalling and eumelanin synthesis models. The two substitutions that define it, norleucine at position 4 and D-phenylalanine at position 7, confer resistance to enzymatic degradation while preserving MC1R affinity. Because it lacks meaningful MC3 and MC4 activity, it is the more selective of the two Melanotan research materials and is often used as the control arm when investigators need to separate MC1R-mediated pigmentation effects from the broader melanocortin activity of Melanotan II.
