Peptide testing in Canada
Every vendor in this market says its material is pure. The claim is free to make and almost nobody who reads it can evaluate it, which is why it carries so little information. This page is about the part that can be evaluated: what the tests actually measure, what the resulting document does and does not establish, and how to check it without taking a seller's word for anything.
The two measurements, and why you need both
Purity and identity are separate questions. Conflating them is the most common way a technically accurate certificate ends up meaning nothing.
HPLC answers “how much”
In reversed-phase high-performance liquid chromatography, the sample is dissolved and driven under pressure through a column packed with a nonpolar stationary phase. Components separate by how strongly each one interacts with that phase, so they emerge at different times, and a detector — usually UV absorbance at 214 nm, where the peptide bond absorbs — records the signal as they come off.
The output is a chromatogram: peaks against retention time. Purity is the target peak's area as a percentage of the total integrated area. Note what that definition contains — it is a share of what was detected, under one method, at one wavelength. It is not a census of everything in the vial.
Mass spectrometry answers “what”
Mass spectrometry ionises the sample and measures mass-to-charge ratio, yielding the molecular mass of what is present. Compared against the mass calculated from the intended amino acid sequence, it establishes identity — that the main component is the molecule named on the label rather than a different peptide that happens to chromatograph similarly.
This is the test that catches substitution and truncation. A sequence missing one residue is a different mass; so is a different compound sold under a more expensive name. HPLC alone would report both as gloriously pure.
The short version: a certificate reporting purity without identity has established that the vial contains 99% of something. Which something is the question you were asking.
Other tests exist and answer other questions again. Endotoxin testing (LAL) addresses bacterial contamination, water content addresses how much of the mass is water, and residual solvent and heavy metal screens address process residues. None of these is implied by a purity figure. Each has to be named on the certificate to have happened.
How to read a Certificate of Analysis
Read the identifying fields before you read the number. The number is the part a forger changes; the identifying fields are the part that makes a forgery checkable. A document missing them is not a weak certificate, it is a picture of one.
Issuing laboratory
Names the party whose reputation is attached to the result, and is the address you send an independent verification request to. A certificate with no laboratory on it cannot be checked by anyone.
Laboratory report ID
The lab's own identifier for the analysis. It is the key that lets the laboratory confirm the report exists. Without it, verification has nothing to look up.
Product and stated identity
What the sample was submitted as. The point of the identity test is to check this claim, so it has to be recorded on the document that reports the test.
Batch or lot number
Ties the analysis to one production run. This is what makes a certificate batch-specific rather than a sample that was once tested well.
Date of analysis
A certificate describes a sample at a point in time. An undated result cannot be aged, and cannot be matched against a production date.
Method
Purity means nothing without the method that produced it. Reversed-phase HPLC at a stated wavelength is a different measurement from an area-percent figure with no method named.
Result, with units
The purity figure and, where identity was tested, the observed and expected mass. A mass that does not match the expected value is the finding, not a rounding error.
What a COA cannot tell you
Four limits, each of which has produced a real failure in this market:
- That the vial in your hand came from the batch described. The certificate documents a sample the laboratory received. Everything connecting that sample to your vial is record-keeping, not chemistry.
- Anything about impurities the method could not see. A UV detector reports what absorbs at its wavelength. Non-absorbing species are simply absent from the chromatogram, and absent is not the same as zero.
- Sterility or endotoxin load, unless those tests are named on the document. Purity and sterility are unrelated properties, and a high-purity powder can carry a high endotoxin burden.
- Its own authenticity. A PDF is trivially editable. This is why the last section of this page exists, and why our own certificates are published as page images rather than source files.
Verifying a certificate independently
Every check described above still runs on documents the vendor handed you. One step does not, and it is worth more than the rest combined: take the report identifier and the laboratory name off the certificate and ask the laboratory.
Laboratories issue reports under their own numbering and can confirm whether a given report exists and what it said. A vendor that has published a real certificate is unaffected by you doing this. A vendor that has published someone else's certificate with the name changed is not. That asymmetry is the whole value of the step.
Where to send the request
Independent peptide purity testing is a cross-border service. The laboratories the research community relies on for this specific work sit in Europe and the United States — Canadian samples travel to them the same way everyone else's do. One consequence worth stating: a vendor advertising “Canadian tested” is making a claim that should be met with two questions — which laboratory, and which report number. Both are answerable in one line by anyone who actually has the document.
Check a certificate you already hold

How this applies to what we sell
Batches are sent to an independent third-party laboratory for HPLC purity analysis, and the result is recorded on a certificate specific to that batch rather than to a representative sample. Our quality assurance page sets out the standard and the process in full.
We publish certificates as rasterised page images rather than source PDFs. Everything a certificate states is also rendered as text, so a reader loses nothing — but a competitor cannot take the file, change the laboratory name and the purity figure, and keep an accredited laboratory's letterhead. Given the fourth limit above, handing out editable certificates would be a strange thing for a testing-first vendor to do.
Batch certificates are published to the certificate archive as they are issued. The verification path above does not depend on us: it works on any vendor's certificate, and it is the one you should run before trusting a purity figure from anyone, ourselves included.
Common questions
How are research peptides tested for purity?+
The standard method is reversed-phase high-performance liquid chromatography. A dissolved sample is pushed through a column that separates its components by how strongly each one sticks to the column packing, and a detector records what comes off and when. Purity is reported as the target peak's share of the total detected peak area, which is why a purity figure is always relative to what the method could detect rather than an absolute statement about the vial.
What is the difference between HPLC and mass spectrometry here?+
They answer two different questions and neither substitutes for the other. HPLC answers 'how much of this sample is the main component' — it quantifies. Mass spectrometry answers 'is the main component the molecule it is supposed to be' — it identifies, by measuring molecular mass to a tolerance fine enough to distinguish closely related sequences. A certificate reporting purity without identity has established that a vial contains 99% of something unnamed.
What does ≥98% purity actually mean?+
That the target peak accounted for at least 98% of the total peak area the detector integrated, under that method, at that wavelength. It does not mean the remaining fraction was identified, and it does not speak to anything the method cannot see. Water content, residual solvents, salt counter-ions and endotoxin are all invisible to a purity-by-HPLC figure and require their own tests.
How do I read a Certificate of Analysis?+
Work through the identifying fields before the numbers. A certificate must name the laboratory, carry a laboratory-issued report identifier, name the product, state the batch or lot it describes, and give the date of analysis and the method used. Then read the result. A document that reports 99% purity but names no laboratory, no report number and no lot is not a certificate of analysis; it is a picture of one.
What can a COA not tell you?+
Four things, and each has produced a market failure. It cannot tell you the vial in your hand belongs to the batch the certificate describes — that link is an administrative claim, not an analytical result. It cannot report on any impurity the method could not detect. It cannot speak to sterility or endotoxin unless those tests are named on it. And it cannot establish its own authenticity: a certificate is a document, and documents are forgeable.
How do I verify a Certificate of Analysis independently?+
Take the report identifier and the laboratory name off the certificate and ask the laboratory, not the vendor. Accredited labs issue reports under their own numbering and can confirm whether a given report exists and what it said. This is the only verification step in the process that does not depend on trusting the party selling you the material, which makes it worth more than every other step combined.
Are there peptide testing labs in Canada?+
Independent purity and identity testing for research peptides is a cross-border service. The laboratories the research community uses most are in Europe and the United States, and Canadian buyers and vendors send samples to them the same way everyone else does. Canadian analytical chemistry capacity certainly exists in academic and contract settings, but the community-recognised names in this specific niche are not Canadian, and a vendor claiming a Canadian testing accreditation should be asked which laboratory and for which report number.
Should a vendor's own lab count as third-party testing?+
No. Third-party means the laboratory has no commercial interest in the result. In-house analysis can be perfectly competent work and it is still the seller marking their own homework. The distinction is not about the quality of the instrument; it is about who bears a cost when the number comes back wrong.
