Long-chain acyl-CoA cannot cross the inner mitochondrial membrane, so beta-oxidation depends entirely on the carnitine shuttle: CPT1 forms acylcarnitine on the outer membrane, a translocase moves it across, and CPT2 regenerates acyl-CoA inside. Carnitine availability is therefore a potential rate limiter on fatty acid oxidation, which is the premise behind most metabolic research using it.
